Agere’s approach to prediction of In Vivo Performance is unique in the industry. Agere uses a combination of proprietary analytical in vitro measurements along with custom in silico models specifically tailored for insoluble compound performance prediction. Factors such as solubility Vs. pH, permeability along the GI tract, partitioning into bile salt micelles, precipitation rates, and the effect of food all have a profound effect on absorption of insoluble compounds. Agere provides this service as a pure modeling effort or as a complete analysis package incorporating critical physiologically relevant in vitro measurement depending on our Customers needs and ultimate desired outcome.

Agere uses both off-the-shelf ACAT and PBPK modeling software along with proprietary algorithms that incorporate the specific requirements of insoluble compounds to perform in-silico in vivo modeling. The output of a typical project can be predictions of fraction absorbed as a function of dose for a variety of animal or clinical models. A typical result is shown below, along with measured in vivo data. Such information is invaluable to selecting the preferred formulation and for designing in vivo studies with formulations of insoluble compounds.
